Pancreatic Enzymes

Pancreatic enzymes are naturally occurring proteins that play an important role in the digestion of food. They effectively hydrolyze a variety of proteins and lipids, using the same natural process as the human digestive system. Trypsin and chymotrypsin are classified as serine proteases with specific cleaving characteristics. Trypsin preferentially hydrolyzes bonds whose carboxyl groups are contributed by lysine (Lys) or arginine (Arg). Chymotrypsin hydrolyzes peptide bonds involving phenylalanine (Phe), tyrosine (Tyr) and tryptophan (Trp). Phospholipase (PA2) is a standardized product, which cleaves the fatty acid at the second position in phospholipids. Neova’s experienced technical and processing advisors work with customers, developing blends and formulating products to suit specific needs. The applications list below serves as a starting point. With the exception of Phospholipase A2, all pancreatic enzymes are lyophilized (freeze-dried), resulting in enhanced solubility and minimal dust.

Egg Proteins

Naturally occurring egg-white proteins are extracted and purified from the hen egg, where they prevent bacterial growth, bind metal ions and vitamins as well as acting as a nutrient source for the developing chicken embryo. Neova uses proprietary extraction and purification processes to produce egg-white proteins for many industries. Avidin is used in immunodiagnostics because of its strong affinity for biotin (vitamin H). Ovalbumin may be used as a very effective and preferable choice over Bovine Serum Albumin in many applications. Likewise, ovotransferrin is very similar to both human and bovine derived transferrins including lactoferrin, and can substitute in many instances. Ovomucoid is a natural trypsin inhibitor. As such, it is commonly used as a replacement for serum trypsin inhibitor in tissue culture research.

Trypsin is an essential enzyme in cell biology, tissue culture, diagnostic and biological research and is widely used as a functional food ingredient due to its versatility in production of hypoallergenic food and nutraceuticals or over-the-counter drugs (OTC). Phospholipase A2 (PA2 also known as PLA2) is a standardized product used in food production as emulsifier or yield ingredient for a vast range of products like cheese, yogurt, bread and baked food. Phospholipase A2 is a major digestive enzyme, among the most studied and it plays a critical role in most physiological processes including the generation of numerous signaling lipids, and in aggregate seems to affect different diseases. For this reason PA2 has application in diagnostics, biological research, vaccines and drug development.

Food Proteins Hydrolysis

The hydrolysis of milk and whey proteins represents an important application area. Hydrolyzing these proteins can enhance functional properties such as whip and taste, increase digestibility and reduce allergenicity. Hydrolysis is also a critical process in food production and has different applications hence it is widely used in food production.

Note: Some proteins contain a natural trypsin inhibitor, such as egg-white and soy protein, making these foods unsuitable for proteolytic enzymes.

Protein Sequencing

Trypsin is commonly used in proteomics for biological research experiments to hydrolyse proteins for mass spectrometry analysis. Trypsin preferentially hydrolyzes bonds whose carboxyl groups are contributed by lysine (Lys) or arginine (Arg). 

Diagnostics

Trypsin has application in diagnosis of disease for its activity in production of polyclonal antibodies that are ideally suited for use in sandwich assays as second stage antigen detectors.

Cell Culture

Trypsin is used to re-suspend cells adherent to the cell culture dish wall during the process of harvesting cells. Some cell types have a tendency to “stick” – or adhere – to the sides and bottom of a dish when cultivated in vitro. Trypsin is used to cleave proteins bonding the culture to the dish, so that the cells can be suspended in fresh solution and transferred to new dishes. Trypsin can also be used to dissociate dissected cells.

 

Raw materials are sourced from government-certified facilities. All extraction and refining is done under cGMP guidelines for active pharmaceutical ingredients (API). Preparations are available in lyophilized (freeze-dried) powder and range in colour from an off white to light beige. These preparations comply with the recommended purity requirements for food grade enzymes provided by the Joint FAO/WHO Expert Committee on Food Additives (JECFA), the Food Chemical Codex (FCC), as well as specific requirements stipulated in the most recent edition of the U.S. Pharmacopoeia. a
